Project description:
Growing evidence suggests that mitochondrial dysfunction and impaired brain energy metabolism may play a role in the pathogenesis of neurodevelopmental psychiatric disorders, including autism spectrum disorder. Our research focuses on the transcription factor TCF7L2, best known for its role in regulating energy metabolism and its association with type 2 diabetes. Recently, mutations in the TCF7L2 gene have been identified as the cause of a rare neurodevelopmental disorder, whose symptoms often include features of autism spectrum disorder.
Our previous studies show that TCF7L2 also regulates energy metabolism directly in the brain. Moreover, its deficiency in the thalamus leads to metabolic changes in thalamocortical circuits and is associated with behavioral deficits. The recruited researcher will investigate the mechanisms underlying these changes: which molecular mechanisms drive them, how interactions between neurons and astrocytes contribute to these processes, and how these changes ultimately affect behavior. The project will combine studies in mouse models with large-scale approaches, biochemical analyses, and behavioral tests.
Key responsibilities include:
Proposing hypotheses, designing and conducting experiments, analyzing results;
Maintaining thorough documentation of work (experiment descriptions, collecting raw data, and visualizing results);
Presenting the project and findings at lab meetings and conferences;
Visualising data, writing research reports and papers;
Assisting less experienced lab members and supervising interns, within mutually agreed scope;
Participating in shared laboratory duties.
